📍 Studying Cyanotoxins in the Alborz Caspian Fish Breeding Pond and its Effect on Fish Tissue
📚 Abstract
Cyanobacteria are one of the ancient microorganisms on the earth that play a significant role in the ecosystem with the ability of photosynthesis and nitrogen fixation. Though, some of these cyanobacteria, by producing dangerous secondary metabolites, are a threat to human and animal health. In this research, initially, samples were taken from the Alborz Caspian fish farm located in the vicinity of the Karaj River, and the resulting samples were cultured and purified in Z8 solid culture medium for morphological identification. Three Calotrix, Nostoc, and Microcystis species were identified in this farm and after gene extraction from these three species, the mcy D gene was recognized with the help of the PCR method. Likewise, to evaluate the effects of eating cyanobacteria on fish tissues, cyanobacterial samples were propagated in a Z8 liquid culture medium and after lyophilization, they were added to the diet of 50 pieces of zebrafish for a period of 30 days. In this test, samples were taken on days 10, 20, and 30 for histopathological examination and measurement of antioxidant enzyme activity. The results attained from the current study designate tissue damage in the intestinal and gill organs and reversible changes in the liver tissue. Besides, a momentous increase in the activity of glutathione peroxidase enzyme in the treatment group compared to the control group on days 20 and 30 and a significant increase in the activity of superoxide dismutase enzyme was observed on day 30. No significant change was observed in catalase enzyme activity.
